Background
BPC-157 was first described in the 1990s by research groups investigating protective fragments of gastric juice protein. Unlike many peptides, it demonstrated notable stability in gastric acid and aqueous solution, which made it a practical candidate for oral and parenteral administration in animal experiments.
Since then it has appeared in several hundred preclinical publications, largely from a small number of research groups, covering gastrointestinal, musculoskeletal, vascular and neurological models.

A separate line of work proposes involvement of the VEGFR2–Akt–eNOS axis. In a rat hind-limb ischaemia model, BPC-157 administration was reported to increase VEGFR2 expression and improve blood-flow recovery, suggesting a pro-angiogenic influence in that setting.
The precise pharmacology, receptor interaction, effective dose range and long-term safety profile of BPC-157 remain unknown. Findings across studies are heterogeneous in species, route, dose and endpoint, which limits direct comparison.
Interpreting the evidence
Readers should note that most published work uses rodent models, small group sizes and short observation windows. Independent replication is limited, and outcomes in animal soft-tissue healing are strongly influenced by loading, rehabilitation and nutrition — variables that are difficult to control.
There is currently no regulatory-approved human dosing protocol for BPC-157. Any laboratory protocol should be defined by the model under study, institutional ethical approval, appropriate controls and predefined endpoints.
